Notes

At this BRCA1 gene SNP position, all 3 missense variants are known: c.301+1G>A, c.301+1G>C and c.301+1G>T. Re c.301+1G>A: Two ClinVar sources classify this mutation as pathogenic/likely pathogenic for breast cancer, however, in a publication () authors associated with a private/commercial laboratory state that it's not likely to result in a significant risk for breast or ovarian cancer, so they classify this mutation as benign. This is also the variant tested on at least some 23andMe DNA chips. Re c.301+1G>C: Annotated as pathogenic and likely to be pathogenic in ClinVar. Re c.301+1G>T: Annotated as likely to be pathogenic by two submitters to ClinVar.
